By Bhavana KunkalikarMar 14 2023Reviewed by Danielle Ellis, B.Sc. In a recent study published in Nature Cell Biology, researchers assessed the association between severe acute respiratory syndrome coronavirus 2 infection and deoxyribonucleic acid damage and cellular senescence.

About the study In the present study, researchers demonstrated that SARS-CoV-2 damages DNA and triggers an altered DNA damage response. Furthermore, reverse transcription-quantitative polymerase chain reaction , immunofluorescence, and immunoblotting were employed to determine the amounts of ribonucleotide reductase M2 messenger RNA and protein. To evaluate if CHK1 depletion is enough to recreate the events after SARS-CoV-2 infection, the researchers investigated the effect of CHK1 depletion by RNA interference.

Infection of human nasal epithelial primary cells with SARS-CoV-2 confirmed DDR activation, as indicated by pRPAS4/8 and H2AX foci. As evaluated by tail moment, DNA fragmentation was induced in both SARS-CoV-2-infected cell lines relative to control conditions.

Flow cytometry demonstrated that cells knocked down for CHK1 aggregated in S-phase. Pulse labeling with BrdU prior to flow cytometry analysis revealed a greater proportion of BrdU-negative S-phase cells in comparison to control samples. The team also noted that the depletion of CHK1 was sufficient to decrease RRM2 levels and result in DNA damage.

Bispecific antibody shows improved efficacy against SARS-CoV-2 variantsBispecific antibody shows improved efficacy against SARS-CoV-2 variantsResearchers from The University of Texas MD Anderson Cancer Center have developed fully human bispecific antibodies, as revealed in a recent study. These antibodies can bind to two distinct epitopes of the receptor binding domain (RBD) of the SARS-CoV-2 spike protein and have demonstrated the ability to neutralize various strains of the virus, including the Omicron variant.
